Ratiometric and colorimetric fluorescent probe for hypochlorite monitor and application for bioimaging in living cells, bacteria and zebrafish

Recently, Shen and Deng group from Wenzhou Medical University have designed and constructed a specific type of hypochlorous acid (ClO-) fluorescent probes with ratiometric and colorimetric response, and successfully applied to bioimage in living cells, bacteria and zebrafish, which were providing important theoretical basis for ROS monitor in organisms and environment.

In previous work (Analyst, 2019; Talanta, 2019; Spectrochimica Acta Part A: Molecular and Biomolecular Spectroscopy, 2020; Analyst, 2020; Dyes and Pigments, 2020), systematic research has been conducted on the application of fluorescent probes. In this work, coumarin-salicylic hydrazide Schiff base (CMSH), a ratiometric and colorimetric fluorescent probe for ClOdetection based on coumarin as the fluorophore unit was rationally designed and synthesized. The results indicated that CMSH exhibits high selectivity and sensitivity for ClOidentification. Additionally, the ratios (I470/I532) displayed brilliant ClO--dependent quick and sensitive performance within 40 s and limitation of 128 nM, respectively. As well as the color of the solution changes from green to colorless accompanied by the fluorescence form green turns into blue with addition of ClO-.
