报告摘要:Antibodies exhibit hunting strategies resembling random walking in initial antibody-patrol stage and confined motion in subsequent antibody-antigen binding stage. When they start to “see” and how they approach each other just before binding remains unknown due to the limitations of detection techniques. Here, utilizing the recently developed method MINFLUX, the antibody starts to “see” the antigen when their distance is about 200 nm. After that, the antibody is rapidly attracted to the antigen through the strategy of targeted motion. Therefore, a new stage, i.e., the antibody-attraction stage, was found which connects the antibody-patrol stage and the antibody-antigen binding stage where strategies with free and confined motions are involved, respectively. Regarding the distance representing the hunting efficiency, our findings elucidate the design of therapeutic antibodies.
